糖尿病大鼠角膜上皮创伤愈合中IGF-1的表达

Delayed corneal wound healing is related to impaired IGF-1 expression in diabetic rats.

摘要: 目的 研究糖尿病大鼠角膜上皮创伤愈合过程中胰岛素样生长因子-1(insulin-like growth factor-1, IGF-1)及胰岛素样生长因子-1受体(insulin-like growth factor-1 receptor, IGF-1R)的表达,从分子水平探讨糖尿病角膜上皮创伤愈合延迟的可能机制。 方法 链脲霉素诱导糖尿病大鼠模型,造模后每周同一时间点监测体质量和空腹血糖变化。制作大鼠角膜上皮创伤模型,于创伤后不同时间点,评价角膜上皮愈合速率;实时定量PCR及免疫荧光检测角膜上皮中IGF-1、IGF-1R的表达。 结果 糖尿病组体质量低于正常组,空腹血糖浓度高于正常组。与正常组相比,糖尿病组角膜上皮愈合速率慢。实时定量PCR及免疫荧光检测显示,新愈合的糖尿病组大鼠角膜上皮中IGF-1、IGF-1R的mRNA、蛋白表达低于正常组。 结论 糖尿病大鼠角膜上皮创伤愈合延迟,IGF-1、IGF-1R表达下调可能是导致愈合延迟的原因之一。

Abstract: Objective To explore the expression of IGF-1 and IGF-1R in corneal wound healing in diabetic rats. Method Male Wistar rats were injected with streptozocin; serum glucose levels and body weight were monitored. Following corneal abrasion, healing rate was evaluated. Real-time PCR and immunofluorescence were used to explore the expression of IGF-1 and IGF-1R. Result Low weights and high glucose levels were observed in diabetic rats. Healing rate was significantly lower in the diabetic rats than in normal rats. mRNA and protein expression of IGF-1 and IGF-1R was significantly lower in the diabetic rat group than in the normal group. Conclusion In diabetic rats, delayed corneal epithelial wound healing might be associated with impaired IGF-1 and IGF-1R expression.
