ARelationship between expression of Survivin and radiosensitization effect of EGCG in xenotransplanted nasopharyngeal carcinoma

Abstract:

To investigate the relationship between expression of Survivin and radiosensitization effects of EGCG in the in vivo growth of nasopharyngeal carcinoma. MethodsThe nasopharyngeal carcinoma undifferentiated cell line (CNE2) was subcutaneously inoculated into 20 nude mice (46weeks, female), and the volume and the shorter diameter of tumors were determined. When the shorter diameter of tumors reached 46?mm, NS, EGCG(50?mg/kg·w), radiotherapy(15?Gy), and EGCG(50?mg/kg·w) followed by radiotherapy(15?Gy) 24 hours later, were administered to the animals. The volume and the shorter diameter of tumors were checked. The excised tumor masses on the 21st day after administration were frozen and processed for reverse transcriptionpolymerase chain reaction(RTPCR) to investigate expression of Survivin mRNR. At the same time, one part of each tumor mass was processed for immunohistochemical TUNEL staining to get the apoptosis index(AI). ResultsEGCG in combination with radiotherapy showed a significant inhibitory effect on both tumor growth and expression of Survivin mRNA(P<0.05). The apoptosis index increased after EGCG treatment in combination with radiotherapy(P<0.05). ConclusionEGCG is a good choice for the treatment of nasopharyngeal carcinoma and has radiosensitization on nasopharyngeal carcinoma. Survivin is a molecular agent taking part in the course of apoptosis induced by EGCG in combination with radiotherapy.
