Expression and  relationship of HMGA1 and Ezrin in laryngeal carcinoma

Abstract:

Objective   To investigate expression of HMGA1 and Ezrin in laryngeal carcinoma and analyze their clinical significance and relationship. Methods   Expression of proteins of HMGA1 and Ezrin was semi-quantitatively analyzed by SP immunohistochemical staining in 50 cases of laryngeal carcinoma. 30 cases of laryngeal polyp and 30 cases of atypical laryngeal hyperplasia were studied as controls. Expressions of mRNA of HMGA1 and Ezrin were analyzed by rea-time PCR in 30 cases of laryngeal carcinoma. Also, 30 cases of  cancer cutting-margin were studied as controls. Results   (1) The positive rates of HMGA1 protein were 68.0%(34/50), 53.0%(16/30) and 13.0%(4/30) in laryngeal carcinoma, atypical hyperplasia of laryngea and laryngeal polyps, while Ezrin protein were 64.0% (32/50), 50.0%(15/30) and 23.3%(7/30), respectively. (2) Expression of HMGA1 and Ezrin proteins and mRNA was closely related to clinical stage, pathological stage and lymph node metastasis (P<0.05), but not  patients′age, gender or tumor location(P>0.05). There was a positive correlation between the protein and mRNA expression of HMGA1 and Ezrin. (r=0.385,P=0.000, by real time PCR;χ2 =7.172,r=0.379,P=0.012, by IHC).  Conclusion   Over-expression of HMGA1 and Ezrin may contribute to the carcinogenesis, development and metastasis of laryngeal carcinoma. They may be new markersto evaluate the prognosis of laryngeal carcinoma.
