TRPV1受体与偏头痛关系的实验研究

Relationship between the TRPV1 receptor and migraine in a nitroglycerin-induced rat model

摘要:

目的    利用硝酸甘油诱导的偏头痛模型,检测瞬时感受器电位香草酸亚1(TRPV1)的表达,探讨TRPV1受体是否参与硝酸甘油诱导的偏头痛过程。方法    成年雌性Wistar 大鼠24只,按随机数字法分为硝酸甘油模型组和对照组。模型组大鼠颈部皮下注射硝酸甘油(10mg/kg),对照组在相同部位注射等量生理盐水。观察大鼠行为表现,分别于注射药物1h、2h、4h和8h取大鼠双侧的三叉神经节及硬脑膜,采用RT-PCR、Western-Blot及间接免疫荧光法检测TRPV1受体表达。结果    对照组各时间点三叉神经节均可见TRPV1受体表达,但各时间点相对表达无显著差异(P>0.05);模型组三叉神经节TRPV1受体1h、2h mRNA和蛋白相对表达量与对照组比较下调,而4h、8h表达升高(P<0.05);模型组三叉神经节TRPV1受体8h表面蛋白相对表达量高于对照组(P<0.05);间接免疫荧光显示模型组硬脑膜8h TRPV1受体表达高于对照组。结论    TRPV1受体在硝酸甘油诱导的偏头痛模型中表达变化,提示抑制或拮抗TRPV1受体将是治疗偏头痛的新思路。

Abstract:

Objective    To investigate the transient receptor potential vanilloid subfamily member 1 (TRPV1) expressed in migraine rat triggered by nitroglycerin so as to explore the possible mechanism of migraine. Methods    24 Wistar rats were randomly divided into the control group and the model group. The migraine rat model (model group) was established after subcutaneous injection of GTN (10mg/kg) in the neck while the control group had saline. After the model was established, the trigeminal ganglion and dura mater were obtained at 1,2,4,8 hours and expressions of mRNA and protein of TRPV1 were detected by RT-PCR, Western-blot and Indirect immunofluorescence. Results    In the control group, the positive reactions of TRPV1 receptor in the trigeminal ganglion were detected at all designated time points, but, no difference among the time points was found (P>0.05). Compared with the control groups, the relative values of mRNA and protein in the trigeminal ganglion in the model groups were lower at 1h and 2h, but higher at 4h and 8h.  The surface protein TRPV1 receptor at 8 h in the trigeminal ganglion in model rats was higher than that in the control groups, and, similar expression was found in the dura mater. Conclusion   The alteration of TRPV1 receptor expression suggests that the TRPV1 receptor may play a role in the pathogenesis of migraine.
