Effect of siRNA by targeting the human hTERT gene on morphology in Hep-2 cells

Abstract:

Objective  To   investigate the effect of the small interfering RNA (siRNA) targeting hTERT gene in Hep-2 cells on morphology. Methods  siRNA targeting site 1718 of hTERT mRNA was designed and synthesized. In vitro cultured Hep-2 cells were transfected with lipofectamine 2000 and the inhibitory effect was observed by phase contrast microscopy and transmission electron microscopy. Results   48 hours after siRNA were transfected into Hep2 cells, numerous round cells and shrinkage of the cellular membrane were observed under phase contrast microscopy, cells showed innocence transformation under HE stain microscopy, and apoptosis and oncosis appeared under transmission electron microscopy. Conclusion  siRNA can promote the Hep-2 cell to show innocence transformation and apoptosis.
