Effect of UVB on glycoprotein nonmetastatic melanoma protein b in human retinal pigment epithelium cells.

摘要:

目的    研究中波紫外(UVB)对RPE细胞GPNMB表达的影响。方法    采用MTT法检测UVB对RPE细胞存活率的影响;实时定量PCR和蛋白印迹法检测UVB对RPE细胞中GPNMB表达的影响。结果    MTT实验表明,经过100μW/cm2光强的UVB辐射处理2h后,RPE细胞会产生明显的影响,其凋亡细胞数目显著增多。RT-PCR、Western blot结果表明,经UVB照射后,RPE细胞中GPNMB的表达显著上升。结论    UVB辐射可抑制RPE细胞的存活率,促进RPE细胞中GPNMB的表达。

Abstract:

Objective    To investigate the effect of ultraviolet b (UVB) on glycoprotein nonmetastatic melanoma protein b (GPNMB) in human retinal pigment epithelium (RPE) cells. Methods    MTT assay was used to determine the effect of UVB on RPE cell survival rate. The effect of UVB on GPNMB in RPE cells was detected by RT-PCR and western blot. Results    MTT assay showed that RPE cells were damaged by UVB irradiation at 100 μW/cm2 for 2 h and the number of apoptotic cells significantly increased. The results of RT-PCR and western blot demonstrated that the expression of GPNMB in RPE cells markedly enhanced after UVB irradiation. Conclusion    UVB irradiation could suppress the survival rate of RPE cells and promote the expression of GPNMB in RPE cells.
