穴位埋线调节变应性鼻炎大鼠鼻黏膜免疫微环境的研究

Catgut implantation at acupoints regulate immunity of sd rat with allergic rhinitis.

摘要: 目的 通过动物实验验证穴位埋线治疗变应性鼻炎(AR)的疗效,并从鼻黏膜免疫微环境方面探讨其作用机制。 方法 将70只健康SPF级SD大鼠随机分为空白组、AR模型组、AR西药组、AR穴位假埋线组、AR穴位埋线组。造模方法采用改进后的卵蛋白注射及其鼻黏膜刺激法。除空白组外各组予以不同治疗方法,治疗末日在卵白蛋白激发后,观察30 min,观察大鼠AR症状,记录并比较各组大鼠鼻部症状评分,之后立即处死动物,采集大鼠鼻中隔黏膜,采用免疫组化方法检测鼻黏膜组织 TGF-β1、IL-17的表达情况。 结果 穴位埋线能明显缓解AR大鼠鼻部症状,如喷嚏、流涕、抓搔鼻部。与模型组比较,穴位埋线能降低鼻黏膜中TGF-β1、IL-17 含量。穴位假埋线也能降低AR大鼠鼻黏膜中TGF-β1含量。 结论 穴位埋线可降低AR大鼠鼻黏膜中TGF-β1、IL-17 含量,从而调节AR大鼠鼻黏膜免疫微环境。

Abstract: Objective To explore the effect of catgut implantation at acupoints on allergic rhinitis(AR)animals and to discuss the mechanism based on pathological morphology and immunology. Methods A total of 70 healthy SPF SD rats were randomly divided into the blank group, the AR model group, the AR western medicine group, the AR false catgut implantation at acupoint group and the AR catgut implantation at acupoint group. AR animal models were established according to the method of improved ovalbumin injection and nasal mucosa stimulation. Every group received different therapies except the blank group. After the last treatment day, ovalbumin nasal mucosa stimulation was carried out, then the symptoms were observed and recorded. SD rats were killed and septum mucosa was collected to detect expressions of tissue TGF-β1 and IL-17 by using immunohistochemical method. Results Catgut implantation at acupoints can obviously alleviate the symptoms of sneezing, runny nose and scratching of AR rats. Compared with model group(Ⅱ), catgut implantation at acupoints can reduce the TGF-β1 and IL-17 content in the AR rat nasal mucosa. False catgut implantation at acupoints can reduce the TGF-β1 content. Conclusion Catgut implantation at acupoints can reduce the TGF-β 1 and IL-17 content to regulate immune system in AR rat nasal mucosa, then alleviate AR rats symptoms.
