STAT3在喉癌中的表达特征及其与Survivin表达的关系

STAT3 expression in laryngeal carcinoma tissues and its relation with Survivin expression

摘要: 目的探讨信号传导与转录激活因子3(STAT3)在喉癌中的表达特征及其与Survivin表达的关系。方法采用免疫组织化学方法检测40例喉鳞状细胞癌组织和12例喉正常黏膜组织中STAT3、磷酸化STAT3(p-STAT3)蛋白和Survivin蛋白的表达;运用逆转录多聚酶链式反应(RT-PCR)检测上述组织中STAT3 mRNA和Survivin mRNA的表达。结合病例资料进行统计学处理,采用χ2、t检验。结果喉癌组织中STAT3、p-STAT3和Survivin蛋白及STAT3 mRNA和Survivin mRNA的阳性表达率均明显高于正常喉黏膜(P<0.01); STAT3、p-STAT3蛋白和STAT3 mRNA在喉癌中的表达均与喉癌的临床分期相关(分别为P<0.05、P<0.01、P<0.01);Survivin蛋白和 Survivin mRNA在喉癌中的表达与临床分期及淋巴结转移相关(P<0.01,P<0.01)STAT3和Survivin的表达呈正相关。结论STAT3的过量表达可能在喉癌的发生、发展中起重要作用,它的异常激活也可能通过上调Survivin基因的表达,延长喉癌细胞生存时间。

Abstract: To investigate the expression characters of signal transducer and activator of transcription 3(STAT3) and its relation with the expression of Survivin in laryngeal carcinoma. MethodsImmunohistochemistry (IHC) was used to determine the protein expressions of STAT3, p-STAT3 and Survivin in 40 laryngeal squamous cell carcinoma tissues and 12 normal control laryngeal mucous tissues. Also RT-PCR was used to determine the expressions of STAT3 mRNA and Survivin mRNA. ResultsThe expressions of STAT3, p-STAT3 and Survivin protein in laryngeal carcinoma tissues were significantly higher than those in normal laryngeal tissues (P<0.01). And the expressions of STAT3 mRNA and Survivin mRNA were significantly higher in laryngeal carcinoma tissues than those in normal laryngeal tissues (P<0.01). Positive expressions of STAT3, p-STAT3 protein and STAT3 mRNA were correlated with the clinical stage and had no relationship with patient age, tumor position, histological grade, and lymph node metastasis, and also positive expression of Survivin protein and Survivin mRNA was correlated with the clinical stage and lymph node metastasis. There were positive correlations between STAT3 and Survivin expression in laryngeal carcinoma. Conclusions Over-expression of STAT3 may play an important role in the genesis and growth of laryngeal carcinoma. STAT3 may prolong the survival time of laryngeal carcinoma by increasing the expression of Survivin.
